Proline will be seeking to complete the job against Uganda Premier league side MYDA FC when the two face off again in Tororo on Friday.
Proline head into this game full of confidence having won 4-1 in the first leg played at Omondi Stadium in Lugogo.
The visitors are expected to ring some changes to their starting team especially in defence with Savio Kabugo earning a place having started on the bench in the first game.
While for the Home side they are welcoming back Batali Moses fully to their starting line team after he sat out the first game due to injury.
MYDA interim coach Elisha Madanda has a huge task to earn his first win at the club against an attack minded side to whom his side lost by three clear goals.
For MYDA to overturn deficit and progress to the next round, they need to score three clear goals without conceding having scored an away goal on Tuesday.
A positive result away from home will see Proline progress to the round of 16 as they seek to defend the title they won in 2019.
Proline are still the defending champions having won the 2018/19 championship when they beat Bright Stars FC in shootouts in a final held at Masaka Recreation ground.
